This Phase 2, open-label study sponsored by Jazz Pharmaceuticals evaluates JZP3507 (ONC206), an oral small-molecule dopamine receptor D2 (DRD2) inhibitor, in adults with recurrent/progressive World Health Organization grade 2–3 meningioma after prior therapy. The primary aim is to characterize antitumor activity alongside safety, tolerability, and pharmacokinetics with continued oral dosing. The target population reflects a high-unmet-need segment where local options (repeat surgery and/or additional radiation) are often exhausted, and systemic therapy choices are not standardized; in practice, many patients cycle through off-label approaches (for example VEGF-pathway agents like bevacizumab, somatostatin analogs, or clinical trials) with modest, inconsistent durability. A key angle is whether CNS-penetrant DRD2 blockade can deliver clinically meaningful disease control in a tumor type where pathway-driven targeted therapies are still emerging and response benchmarks are generally low.
