Phase 1 CART-EGFR-IL13Rα2 Cells Targeting EGFR and IL13Rα2 in GBM

Trial ID
NCT06973096 on ClinicalTrials.gov
Cancer type
Glioblastoma
Stage of research
First-in-human
Location
University of Pennsylvania
Published in
New in Oncology - Aug 29, 2025

This open-label study at the University of Pennsylvania examines newly diagnosed glioblastoma patients with EGFR-amplification and MGMT-unmethylation post-radiotherapy. Eligible patients undergo a two-step screening, followed by apheresis and placement of a CSF-ventricular reservoir for intracerebroventricular delivery of fixed doses of CART-EGFR-IL13Rα2 cells—engineered T cells with dual CARs targeting a cryptic EGFR epitope 806 and IL13Rα2. This first-in-human investigation explores a novel intrathecal CAR T cell strategy for high-risk GBM. Current SoC after chemoradiation is maintenance temozolomide (± Tumor Treating Fields) with limited durability; localized CAR-T delivery aims to improve control where systemic options are few.
