In this phase 1 trial, D3S-001 was evaluated in KRAS G12C-mutant advanced tumors (42 patients in the dose-escalation phase and 20 NSCLC patients pretreated with prior G12C inhibitors) and exhibited dose-dependent pharmacokinetics with no dose-limiting toxicities. An overall objective response rate of 73.5% was seen in G12C inhibitor-naïve patients across NSCLC, colorectal, and pancreatic ductal adenocarcinoma. Even in G12C inhibitor-pretreated NSCLC, a 30% objective response rate with an 80% disease control rate was achieved.
Overall, D3S-001 shows a favorable safety profile and promising efficacy that may help overcome resistance in KRAS G12C tumors, supporting further clinical development, especially for patients progressing after earlier G12C inhibitor therapy.
