Advenchen Pharmaceuticals’ open-label Phase 1b/2a study evaluates catequentinib hydrochloride (AL3818, also known as anlotinib), a multikinase inhibitor of angiogenesis-related signaling including VEGFR, FGFR, and PDGFR pathways. The program examines AL3818 alone and combinations that include the PI3K/mTOR inhibitor AL58805, with temozolomide or lomustine-containing strategies specified for glioblastoma.
Dose escalation is followed by disease-specific expansion in advanced or recurrent endometrial cancer, low-grade serous ovarian cancer, soft tissue sarcoma, and glioblastoma. Endpoints include tumor response for non-CNS cohorts and 6-month progression-free survival in the glioblastoma cohort. These disease-specific cohorts explore different treatment backbones and clinical needs; activity and tolerability must be interpreted within each cohort rather than pooled across unrelated tumor types. The key issue is whether the selected combination improves disease control with tolerable toxicity in each individual cohort.
