This first-in-human, open-label, multicenter Phase 1a/1b dose-escalation/expansion study evaluates ARR-002 in adults with locally advanced or metastatic ovarian or endometrial cancer. ARR-002 (ArriVent BioPharma) is a tetravalent dual-target antibody-drug conjugate that binds MUC16 and NaPi2b to drive tumor-cell internalization and deliver the cleavable-linker payload vcMMAE (an auristatin microtubule inhibitor), aiming to improve payload delivery across heterogeneous antigen expression versus single-target ADCs. The trial’s primary focus is safety/tolerability with PK and immunogenicity, plus preliminary antitumor activity to select dose/schedule for expansion. The target population includes patients with few effective options after standard therapies; today’s care for platinum-resistant ovarian cancer is sequential systemic treatment (single-agent chemo, bevacizumab combinations in selected patients, and FRα-targeted ADC therapy when eligible), while recurrent endometrial cancer commonly uses chemo and, depending on biomarkers, immunotherapy approaches—this trial tests an ADC strategy in a setting where responses are often short-lived.
