NextPoint Therapeutics’ first-in-human, open-label Phase 1 study dose-escalates NPX372, an IgG-like bispecific antibody that binds B7‑H7 (also called HHLA2/B7‑H7 axis target) on tumor cells and CD3 on T cells to redirect cytotoxic T cells toward B7‑H7–expressing cancers. Patients with advanced/metastatic, relapsed/refractory colorectal cancer are included, along with other selected solid tumors such as NSCLC, renal cell carcinoma, pancreatic ductal adenocarcinoma, ovarian cancer, biliary tract cancers, and gastric/gastroesophageal cancers, with dose escalation followed by potential expansion once a tolerable dose is identified. The key angle is tackling solid tumors with a CD3 engager while aiming for target-dependent activity (a common challenge for T‑cell engagers in solid tumors is therapeutic window/cytokine toxicity). Today, these populations typically cycle through sequential systemic therapies—chemotherapy backbones (fluoropyrimidine/platinum/irinotecan), targeted agents when biomarkers fit (e.g., EGFR/BRAF/HER2/FGFR2/IDH1 subsets), and immunotherapy mainly in MSI‑H/dMMR CRC or selected PD‑L1–positive tumors—so this trial tests whether B7‑H7–directed T‑cell redirection can open an “immune-responsive” path for patients who are often IO-nonresponsive.
