This Phase 1a/1b, disease-specific basket study tests BMS-986504 (navlimetostat), an MTA-cooperative PRMT5 inhibitor designed to exploit the MTAP-deletion/MTA accumulation vulnerability, in combination with standard regimens across multiple metastatic or unresectable MTAP-deleted solid tumors. Memorial Sloan Kettering Cancer Center sponsors the trial, while Bristol Myers Squibb owns BMS-986504; the key goal is to establish safety/tolerability and a recommended combination dose while looking for early efficacy signals in defined tumor cohorts. Interventions pair BMS-986504 with established Bristol Myers Squibb immunotherapy in diffuse pleural mesothelioma (nivolumab, a PD-1 antibody, plus ipilimumab, a CTLA-4 antibody), with nivolumab + FOLFOX chemotherapy in PD-L1 CPS>1 gastroesophageal cancer, and with gemcitabine plus platinum (cisplatin or carboplatin) in later-line urothelial carcinoma followed by BMS-986504 maintenance. Today’s care is histology- and line-specific (e.g., nivo/ipi is a common first-line option in unresectable pleural mesothelioma; first-line gastroesophageal cancer often uses platinum/fluoropyrimidine chemo plus PD-1 therapy when PD-L1 positive; metastatic urothelial cancer typically uses platinum-based chemo then checkpoint inhibitors and/or ADCs), and this trial asks whether adding a precision PRMT5 strategy can deepen responses in the MTAP-deleted subset (MTAP loss is a recurring alteration across many solid tumors, often alongside CDKN2A).
