DualityBio’s first-in-human DB-1317 is Q3W, with accelerated titration/BOIN escalation then expansion in GI-heavy tumors (gastric, CRC, PDAC). GI cancers still depend on chemo with few durable targets; ADAM9 overexpression (especially in renal cell carcinoma, breast cancer, liver cancer, prostate cancer, lung cancer, gastric cancer, and cervical cancer) makes a credible ADC target if exposure and bystander effect translate. Safety/PK, ORR, and biomarker-enrichment readouts will decide where to focus.
