MSK identifies the maximum safe dose of on-site–manufactured CD19 CAR-T for primary/secondary CNS lymphoma (incl. DLBCL), using CliniMACS Prodigy (Miltenyi Biomedicine), leukapheresis, conditioning chemo, and infusion with serial CSF/blood sampling. HD-MTX ± rituximab and transplant can fail in CNS disease; adapting CAR-T to CNS aims for deeper remissions where systemic options falter. Neurotoxicity monitoring and early response signals will drive next steps.
