This multicenter, open-label, multiple-dose first-in-human trial by VelaVigo Bio Inc evaluates VBC101 using an accelerated titration in Phase 1 dose escalation with a BOIN design and backfill cohorts to define the maximum tolerated dose and recommended Phase 2 dose. In Phase 2a, dose optimization and cohort expansion assess safety, tolerability, and preliminary efficacy in participants with advanced/metastatic solid tumors lacking standard-of-care options; single-antigen ADCs can fail via antigen loss—dual-targeting EGFR+cMET aims to blunt escape in heterogeneous tumors, including Lung Adenocarcinoma, Colorectal Cancer, Glioblastoma, Laryngeal Squamous Cell Carcinoma, and Chordoma, but best to have mass spec protein analysis whenever possible.
