This global open-label study evaluates the safety, pharmacokinetics, pharmacodynamics, immunogenicity, and efficacy of AZD4512 administered as monotherapy dose escalation (with backfill cohorts) or in combination with other anticancer agents. It enrolls patients with relapsed or refractory B-cell non-Hodgkin lymphoma—including large B-cell, follicular, and mantle cell subtypes—many of whom have received prior anti-CD20 (and in some cases BTKi) therapy to establish the optimal biological and maximum tolerated doses. In the post-CD19 CAR-T/bispecific era, CD22-directed ADCs offer a chemo-free cytotoxic strategy when anti-CD20-based options have been exhausted.
