AbbVie is running this Phase 2, open-label, randomized master protocol (M25-709; sometimes listed as FLORENZA/GOG-3126) to optimize mirvetuximab soravtansine (MIRV), a folate receptor alpha (FOLR1/FRα)–directed antibody-drug conjugate carrying the DM4 microtubule inhibitor payload, across multiple combination backbones in FRα-positive ovarian/fallopian tube/primary peritoneal cancer. Participants are assigned to substudies comparing two MIRV doses in combination with bevacizumab (Genentech/Roche; anti-VEGF monoclonal antibody), carboplatin (platinum DNA crosslinker), or triple therapy (MIRV + bevacizumab + carboplatin), with some arms transitioning to MIRV (± bevacizumab) maintenance. Eligibility centers on FRα positivity by the Ventana FOLR1 assay and high-grade serous ovarian/fallopian tube/primary peritoneal cancer across frontline, second-line, and relapsed settings depending on substudy. Scientifically, the angle is practical and commercial: MIRV is already an FRα-directed platform, so the study focuses on dose/partner optimization to improve benefit–toxicity in common real-world regimens. Today’s care typically uses platinum-taxane chemotherapy in first line with bevacizumab and/or PARP inhibitor maintenance when appropriate, and in recurrence relies on platinum rechallenge when sensitive or sequential non-platinum chemo ± bevacizumab and other targeted options when eligible.
