Acimtamig (AFM13) is a tetravalent CD30/CD16 innate-cell engager that redirects NK cells; AlloNK (AB-101) is an off-the-shelf cord-blood NK-cell product co-infused with IL-2. In the completed dose-finding stage of the open-label phase 2 LuminICE-203 trial (NCT05883449), 22 adults with relapsed/refractory cHL (classical Hodgkin lymphoma) after ≥2 prior lines—including brentuximab vedotin and PD-1 blockade (median 5 prior lines)—received acimtamig 200–300 mg plus escalating AlloNK doses over three 58-day cycles. Independent review showed an 86% ORR and with 55% CR; 68% of responses were ongoing at December 2024 cutoff. No graft-versus-host disease, ICANS, or ≥grade 3 CRS occurred, and cytokine-related events were grade ≤2. By comparison, pembrolizumab monotherapy yields ~69% ORR and 22% CR in the KEYNOTE-087 post-BV/PD-1 population, highlighting deeper remissions with this NK-engager combo.

Take home

If these CR-heavy results persist in larger cohorts, the plug-and-play acimtamig + AlloNK regimen could offer CAR-like efficacy without bespoke manufacturing—potentially redefining salvage therapy for post-BV/PD-1 Hodgkin patients.
