MK-1084 (KRAS G12C inhibitor) plus cetuximab drives 50% responses in advanced colorectal cancer

Trial ID
NCT05067283 on ClinicalTrials.gov
Cancer type
Colorectal Cancer
Published in
New in Oncology - May 29, 2025

Phase 1 KANDLELIT-001 (NCT05067283) enrolled pre-treated KRAS G12C-mut CRC: 53 received MK-1084 monotherapy (ORR 36%), 34 got MK-1084 + cetuximab after ≥2 prior lines (ORR 50%), and 20 front-line patients received MK-1084 + cetuximab + mFOLFOX6 (ORR 14%); all responses were partial. Grade ≥3 treatment-related AEs occurred in 9%, 18%, and 25%, respectively (rash/dermatitis and nausea most common), with one DLT (febrile neutropenia) and no treatment-related deaths.

Take home

Early activity suggests MK-1084 could outshine first-generation KRAS G12C regimens in CRC, but durability—and whether dermatologic toxicity stays manageable—are open questions.
