Phase 1/1b CHAMP adds CCR5 blockade with leronlimab to surgery, hepatic arterial floxuridine, and systemic therapy for colorectal liver metastases

City of Hope’s single-arm CHAMP study evaluates leronlimab, CytoDyn’s CCR5-blocking monoclonal antibody, with a liver-directed treatment program for resectable, liver-dominant colorectal cancer metastases. Participants receive weekly subcutaneous leronlimab with systemic therapy before liver surgery and hepatic arterial infusion (HAI) pump placement. After postoperative recovery, protocol treatment adds HAI floxuridine, a fluoropyrimidine antimetabolite, and resumes leronlimab with systemic therapy.

The Phase 1/1b focus is safety and feasibility of combining CCR5-related immune modulation with regional chemotherapy and surgery. HAI is a specialized liver-directed approach rather than a replacement for systemic management of extrahepatic disease. The study explores whether this combination can improve a perioperative treatment strategy, but any response or recurrence signal will need to be interpreted in the context of patient selection, surgery, and the other active treatments. The registry currently lists this study as not yet recruiting.
