Linvoseltamab is an off-the-shelf BCMA×CD3 T-cell–engaging bispecific antibody compared here with elranatamab, another BCMA×CD3 bispecific, using an unanchored matching-adjusted indirect comparison across LINKER-MM1 (NCT03761108; BCMA-therapy–naïve triple-class–exposed RRMM; n=107; median follow-up 21.3 mo) and MagnetisMM-3 Cohort A (NCT04649359; n=123; median follow-up 33.9 mo). After matching, linvoseltamab showed higher ORR (71.5% vs 61.0%; p<0.05) and ≥CR rate (50.5% vs 37.4%; p<0.05), with numerically favorable DOR (HR 0.82), PFS (HR 0.86), and OS (HR 0.67; p=0.08); OS also favored linvoseltamab by restricted mean survival time (+3.47 mo; p=0.04). Against the broader BCMA-bispecific benchmark, teclistamab in MajesTEC-1 produced ORR ~63% and ≥CR ~39%, so linvoseltamab’s adjusted response depth sits above the usual class ballpark, though no matched safety comparison was reported.
Take home
Linvoseltamab looks competitive within the crowded BCMA-bispecific lane, especially on depth of response, but this is still an unanchored cross-trial comparison—not a head-to-head trial—so safety, infection burden, dosing logistics, and randomized data remain the real differentiators.
