IMNN-001 works by stimulating local production of IL-12, which activates immune cells to attack the cancer. In the randomized phase I/II OVATION-2 trial (NCT03393884), 112 women with stage III/IV epithelial ovarian cancer received standard carboplatin-paclitaxel with or without weekly intraperitoneal IMNN-001 during neoadjuvant and adjuvant cycles; median overall survival rose to 46.0 vs 33.0 months (HR 0.69) and progression-free survival to 14.9 vs 11.9 months (HR 0.79), with no cytokine-release syndrome and only grade 1-2 abdominal symptoms added.

Take home

If phase III confirms these survival gains, IMNN-001 could usher in the first intraperitoneal gene-immunotherapy to meaningfully boost front-line outcomes—especially valuable for HRD-positive patients poised for PARP maintenance (based on subgroup analysis).
