In the CALGB/SWOG 80405 trial, high tumor miR-494 levels were significantly associated with shorter progression‐free survival overall and in cetuximab‐treated metastatic colorectal cancer patients, while in vitro modulation of miR-494 in HCT116 and HT29 cells showed that its inhibition led to reduced PAK1 and increased CDH1 expression, affecting cell colony formation.
These data suggest that miR-494 not only serves as a potential prognostic biomarker in mCRC—especially in the context of EGFR-targeted therapy—but also underscores the need to further explore miR-494–driven pathways for novel therapeutic interventions.
