In this first-in-human Phase 1 study of ANK-101—an IL-12 conjugated to aluminum hydroxide—administered in a dose-escalation design in heavily pretreated patients with accessible advanced solid tumors, the treatment was well tolerated with no grade ≥3 adverse events, low systemic exposure, and demonstrated biological activity including increases in serum IFN-γ, upregulation of PD-L1, and enhanced CD8+ T cell infiltration, yielding a disease control rate of 63.6% (1 partial response and 7 stable diseases).
The findings suggest that ANK-101 can safely harness localized IL-12-driven immune activation to achieve durable disease control, offering a promising new immunotherapeutic strategy for patients with advanced refractory tumors.
