ZG006 is a unique designed T cell engager, targeting CD3 and Delta-like ligand 3 (DLL3) on DLL3-expressing tumor cells such as small cell lung cancer (SCLC) or neuroendocrine carcinoma (NEC). In the phase 1 study with 45 patients (23 evaluable), ZG006 monotherapy achieved a 60.9% ORR (with 66.7% ORR in low/medium DLL3 expressors) and 78.3% DCR in SCLC/NEC, with manageable cytokine release syndrome and a single DLT at 100 mg; drug exposure increased nearly dose-proportionally with a ~4‑day half‐life. Eleven pts (24.4%) experienced serious TRAEs with no discontinuations for AEs.
Take home
ZG006’s CD3/DLL3 T cell engager joins the ‘DLL3 is a success at ASCO 2025’ story with a nice 60% ORR in SCLC/NEC. It’s a bit too early to say if it will emerge as the leading DLL3 immunotherapy, but the numbers look strong and the tolerability looks good too.
