In this early-phase, single-arm trial, cisplatin-ineligible patients with locally advanced head and neck squamous cell carcinoma received definitive radiation therapy alongside tolinapant, an IAP antagonist, with manageable toxicities and encouraging disease control at a median follow-up of 12.4 months; notably, flow cytometry revealed significant increases in activated CD8+ T cells in half of the patients.
The regimen not only proved safe and feasible but also hints at an enhanced anti-tumor immune response, supporting further exploration of tolinapant plus radiation as a promising platinum-free treatment strategy.
