In a Phase I study of relapsed/refractory AML, SENTI-202—a logic gated CAR NK cell therapy targeting both CD33 and FLT3 while sparing EMCN+ healthy HSPCs—demonstrated marked blast reduction with MRD-negative complete remissions in responders, along with a >10-fold decrease in leukemic stem cells and evidence of normal hematopoietic recovery in PB and BM.
These correlative data highlight SENTI-202’s innovative dual mechanism that selectively eradicates AML cells while protecting normal hematopoiesis, offering a promising off-the-shelf immunotherapy approach for hematologic malignancies.
