In the AK105-304 Phase 3 trial, penpulimab combined with gemcitabine and cisplatin/carboplatin showed a statistically significant improvement in progression‑free survival (9.6 vs. 7.0 months; HR=0.45) versus placebo with chemotherapy, and longer duration of response, while overall survival benefits became more evident after adjusting for crossover, all with a manageable safety profile amid a diverse, global patient population.
The study establishes penpulimab plus chemotherapy as a promising new first‑line treatment option for recurrent or metastatic nasopharyngeal carcinoma on a global scale, potentially altering the therapeutic landscape for this challenging disease.
