This Phase 1, multicenter, open-label study from Juno Therapeutics, a Bristol Myers Squibb company, evaluates BMS-986527, a healthy-donor-derived, allogeneic CD70-targeted CAR T-cell therapy in unresectable or metastatic renal cell carcinoma with a clear cell component. Nonrandomized cohorts examine CAR T cells alone, with nivolumab, or with cabozantinib. These partners respectively block PD-1 and inhibit multiple kinases including MET, AXL, and VEGFR2.

Prior-treatment requirements differ: the monotherapy and nivolumab cohorts allow specified prior standard immunotherapy and/or TKI exposure or documented inability to receive those treatments, while the cabozantinib cohort has distinct restrictions including no previous TKI treatment. The goals are safety, dose selection, and early activity. The study tests whether an off-the-shelf cellular therapy can produce useful disease control in a solid tumor with frequent CD70 expression. CD70 expression supplies a rationale, not proof of tumor specificity or protection from CAR T-cell, lymphodepletion, or combination-related toxicities.
