Phase 2 ADT (GnRH antagonist/agonist) + ultra-hypofractionated SBRT in ARTERA-low high-risk prostate cancer

Trial ID
NCT07801664 on ClinicalTrials.gov
Cancer type
Prostate Cancer
Stage of research
Exploratory
Location
New York, NY (single-site, NYU Langone Health)
Published in

NYU Langone Health is running this single-arm, open-label Phase 2 trial testing whether short-course androgen deprivation therapy (ADT) for 6 months can be paired with dose-escalated radiotherapy (ultra-hypofractionated SBRT to prostate ± brachytherapy boost, with treatment to prostate and nodes) in NCCN high-risk localized prostate cancer patients who have low or intermediate-risk ARTERA AI scores. ADT is delivered using relugolix (an oral GnRH receptor antagonist; Sumitomo Pharma/Myovant Sciences, now Pfizer), degarelix (injectable GnRH receptor antagonist; Ferring), or leuprolide (GnRH receptor agonist; marketed by multiple companies). The scientific angle is biomarker-informed de-intensification: selecting “biologically lower-risk” high-risk patients by ARTERA to see if outcomes remain strong with shorter ADT exposure. Today’s care for high-risk localized disease commonly includes definitive radiation to prostate ± pelvic nodes plus longer-course ADT (often 18–36 months), and sometimes systemic intensification in selected patients; this study asks if a shorter, more tolerable hormonal course is enough for an ARTERA-selected subset.
