AbbVie’s AndroMETa-118 (NCT07769502; AbbVie study M26-118) is a Phase 2, open-label master protocol in pancreatobiliary cancers testing telisotuzumab adizutecan (ABBV‑400), a c‑Met/HGFR–directed monoclonal antibody–drug conjugate that delivers a topoisomerase inhibitor payload to c‑Met–expressing tumor cells. Substudy 1 includes an initial dose-escalation to define doses for optimization, followed by a randomized dose-optimization stage comparing two ABBV‑400 dose levels plus gemcitabine versus standard gemcitabine + nab‑paclitaxel. Gemcitabine is an antimetabolite chemotherapy (nucleoside analog) and nab‑paclitaxel is an albumin-bound microtubule inhibitor; the combination is a common first-line regimen for metastatic pancreatic ductal adenocarcinoma and related pancreatobiliary cancers, alongside alternatives like modified FOLFIRINOX in fit patients. The core scientific/commercial angle is whether a c‑Met–targeted, topo‑payload ADC can improve outcomes beyond chemo backbones in a setting where incremental gains are hard-won, while the master-protocol structure allows AbbVie to add/iterate combination substudies efficiently.
