AbbVie’s Phase 3, multicenter, randomized, open-label study tests whether adding etentamig—an anti-BCMA×anti-CD3 bispecific antibody that redirects T cells to kill BCMA-expressing myeloma cells—improves outcomes when combined with pomalidomide, an IMiD (cereblon E3 ligase modulator) with direct anti–plasma cell and immune-stimulatory effects. The trial starts with a safety run-in (etentamig + pomalidomide) and then randomizes ~520 adults with relapsed/refractory multiple myeloma after 1–3 prior lines (must have had lenalidomide) to etentamig + pomalidomide versus investigator’s choice “standard available therapy.” Control options include daratumumab (CD38 mAb) + pomalidomide + dexamethasone (DPd), daratumumab + carfilzomib (proteasome inhibitor) + dexamethasone (DKd), or teclistamab (BCMA×CD3 bispecific) monotherapy. The commercial/scientific angle is a registrational head-to-head that positions a next-generation BCMA bispecific (engineered for potentially improved tolerability and dosing convenience) earlier in the treatment sequence, where current care commonly relies on anti-CD38 combinations, proteasome-inhibitor triplets, and increasingly BCMA-directed T-cell redirection in later lines.
