This Phase 1, first-in-human, open-label dose-escalation with optional expansion study evaluates FX-111 in adult men with metastatic castration-resistant prostate adenocarcinoma that has progressed on androgen-deprivation therapy. Patients take once-daily oral FX-111 across multiple dose levels to define safety/tolerability, characterize pharmacokinetics/pharmacodynamics, and look for early anti-tumor signals (including disease control and progression endpoints). FX-111 (Flare Therapeutics) is a small-molecule targeted protein degrader designed to selectively eliminate ARON—the transcriptionally active, hormone-bound androgen receptor—aiming to suppress AR-driven gene programs even when tumors have adapted to prior AR pathway inhibitors. The key angle is mechanistic differentiation within the crowded AR space: rather than only antagonizing the receptor, it attempts to remove the active AR complex that drives transcription. Today’s care for mCRPC is continued ADT plus sequential AR pathway inhibitors (e.g., abiraterone or enzalutamide), taxane chemotherapy, and biomarker-directed options (e.g., PARP inhibitors for HRR-mutated disease) as disease and prior treatments dictate.
