This first-in-human, open-label, multicenter Phase 1/2 study evaluates ANS02 monotherapy in adults with locally advanced or metastatic EGFR mutation–positive non-small cell lung cancer (NSCLC), focusing on safety/tolerability, pharmacokinetics, and preliminary anti-tumor activity. ANS02 is being developed by Avistone Biotechnology (Beijing Avistone) as a “4th‑generation EGFR-TKI,” i.e., a small-molecule inhibitor designed to block oncogenic EGFR signaling; Avistone’s public materials do not specify the exact mutant selectivity/resistance-mutation profile, so the key scientific question is whether it can extend benefit after prior EGFR TKIs while maintaining a tolerable therapeutic window. The trial’s dose-escalation and expansion structure is intended to establish a recommended dose and then look for early signals of response in molecularly defined NSCLC. In this setting, today’s care is frontline osimertinib for common activating EGFR mutations, with later-line options guided by resistance mechanisms (platinum doublet chemotherapy, amivantamab-based strategies in select contexts, and clinical trials), making a next-generation EGFR inhibitor commercially interesting if it can address common resistance patterns (and ideally CNS disease).
