This Phase 2, open-label, multicenter, single-arm study (no comparator) evaluates inobrodib in combination with pomalidomide and dexamethasone (often described as “InoPd”) in heavily pretreated relapsed/refractory multiple myeloma. Inobrodib (CellCentric Ltd.) is an oral, first-in-class p300/CBP bromodomain inhibitor intended to disrupt transcriptional programs that myeloma cells depend on. Pomalidomide (a Cereblon-binding IMiD from Bristol Myers Squibb/Celgene) promotes degradation of Ikaros/Aiolos and augments immune-mediated anti-myeloma activity, while dexamethasone adds lympholytic steroid activity and symptom control. The scientific/commercial angle is an all-oral, novel epigenetic backbone layered onto a familiar IMiD+steroid platform, aiming to deliver meaningful activity in patients who have exhausted multiple prior classes. Today’s care for R/R myeloma is sequential “triplet” or “quad” regimens (often anchored by anti-CD38 antibodies, proteasome inhibitors, and/or IMiDs), followed by BCMA- or GPRC5D-directed therapies (bispecifics or CAR-T) when eligible; this program targets an epigenetic vulnerability as an alternative/add-on path after standard classes.
