This trial evaluates safety, tolerability, pharmacokinetics, and early anti-tumor activity of perioperative combination treatments in patients with locally advanced resectable gastric, gastroesophageal junction, or esophageal adenocarcinoma. Interventions include AZD0901—a CLDN18.2-directed MMAE toxin inhibitor ADC—and rilvegostomig, a Tigit/PD-1 bispecific mAb, alongside chemotherapy (5-FU, capecitabine, or FLOT) and trastuzumab deruxtecan (a HER2-directed ADC with a DXd payload), using a master protocol design sponsored by AstraZeneca.
