This Phase 3 randomized, open-label trial at GlaxoSmithKline compares belantamab mafodotin (a BCMA-directed antibody-drug conjugate with a tubulin inhibitor payload) plus lenalidomide and dexamethasone versus daratumumab (a CD38-directed monoclonal antibody) with lenalidomide and dexamethasone. The study enrolls newly diagnosed multiple myeloma patients ineligible for transplant, aiming to improve progression-free survival and minimal residual disease status. D-Rd is a widely used standard backbone for transplant-ineligible NDMM; this trial asks if moving a BCMA-ADC earlier can beat D-Rd on PFS/MRD while managing ocular toxicity.
