Marker Therapeutics’ PANACEA Phase 1 dose-escalation with expansion tests MT-601, an autologous (patient-derived) multi–tumor-associated antigen (multiTAA)–specific T-cell product designed to recognize several shared tumor antigens rather than a single target. Patients undergo apheresis to manufacture MT-601, then receive escalating cell doses (200M, 400M, 800M, 1200M cells) administered during the “off week” of an ongoing chemotherapy schedule, followed by a dose-expansion cohort at the recommended dose. The primary focus is safety/tolerability, with preliminary efficacy endpoints including overall response rate and duration of response by standard imaging criteria. The eligible population is locally advanced unresectable or metastatic pancreatic cancer that is not progressing on prior FOLFIRINOX or nal-IRI–based regimens (FFX/NLX). This is a manufacturing- and logistics-intensive, personalized T-cell strategy in a disease where today’s care is sequential multi-agent chemotherapy (often FOLFIRINOX or gemcitabine/nab-paclitaxel first-line, then nal-IRI/5-FU–based regimens) with limited durable control, so the key angle is whether a multi-antigen cellular immunotherapy can add incremental benefit without disrupting chemotherapy delivery.
