This Phase 1 study led by the National Cancer Institute assesses safety, tolerability, optimal dosing, and early efficacy of oral CA-4948 combined with intravenous pembrolizumab in patients with metastatic urothelial cancer resistant to PD-1/PD-L1 therapy. CA-4948 is a small molecule kinase inhibitor targeting IRAK4/FLT3, while pembrolizumab is a PD-1–directed antibody; integrated imaging and biopsy evaluations will explore this novel combination strategy.
