WU-CART-007 is a fratricide-resistant, healthy-donor CAR-T that targets CD7. In the phase 1/2 dose-escalation/expansion trial (NCT04984356), 28 heavily pre-treated patients received a single infusion after standard or “enhanced” lymphodepletion. For 11 evaluable patientsat the RP2D of 9 × 10^8 cells plus enhanced LD, overall response reached 90.9% with a 72.7% composite CR rate, including one IEC-HLH case but no ≥G3 neurotoxicity or GvHD. CRS was common (88.5%; 19% ≥G3). By comparison, salvage nelarabine shows ~50% ORR and 36% CR in this setting, highlighting WU-CART-007’s step-change in depth of response.
Take home
Off-the-shelf CD7 CAR-T could finally give R/R T-ALL/LBL a high-response option, provided CRS remains manageable and durability matches the early fireworks.
