This phase 2 trial (NCT03779191) with 41 patients combines alectinib with bevacizumab in advanced ALK-positive non-small cell lung cancer (NSCLC). The combination achieved a 12-month PFS rate of 97.1% (95% CI 92.6–100), 36-month OS of 87.9%, and ORR of 100%. Discontinuation of bevacizumab was observed in 18 patients (43.9%) due to proteinuria-related events.
Take home
The results suggest that combination treatment significantly improves outcomes with first-line therapy for ALK-rearranged NSCLC, but with very high discontinuation rates, it’s unlikely that this will become a standard of care combination.
