An indirect treatment comparison using matched patient-level data from KEYNOTE-B10 and six pivotal trials revealed that pembrolizumab plus carboplatin and paclitaxel (KN-B10) delivers comparable or superior objective responses, overall survival, and progression-free survival relative to established regimens—including EXTREME, TPEx, and KN-048—especially demonstrating improved outcomes versus platinum plus 5-FU.
This analysis supports the use of KN-B10 as an effective first-line alternative for recurrent/metastatic HNSCC, highlighting its potential to enhance clinical outcomes while providing a vital option amid evolving immuno-oncology strategies.
