A phase escalation study in bone-predominant metastatic castration-resistant prostate cancer (CRPC) demonstrated that a dose-dense regimen of docetaxel combined with standard radium-223—with a lead-in cycle and G-CSF support—produced encouraging PSA responses (51.4% achieving PSA50; 25.7% achieving PSA90), an 11.7-month median progression-free survival, and a 20.1-month median overall survival, all without febrile neutropenia.
This approach mitigates myelosuppression and offers a viable, enhanced treatment strategy that may extend to high-risk metastatic settings when combined with hormone intensification.
