In the Phase III TOPAZ-1 study of patients with advanced biliary tract cancer, a combination of durvalumab and gem/cis resulted in a median OS of 12.9 months, compared to 11.3 months for the placebo with gem/cis, after a median follow-up of 41.3 months. Among participants who achieved disease control, the 36-month OS rate was higher in the durvalumab plus gem/cis group (17%) versus the placebo plus gem/cis group (7.6%). Serious adverse events were comparable between the treatment arms.
Take home
Though incremental, any gain in HCC is a welcome one, and with the 36-mo OS, perhaps we smell a long-tail just like the CTLA-4 melanoma days… Perhaps a bispecific will follow.
