In this phase 1 trial of 70 heavily pretreated patients with HER2-expressing gastroesophageal adenocarcinoma, zanidatamab monotherapy yielded a confirmed objective response rate of 32.1%, while combining it with chemotherapy increased the ORR to 48.6%, with manageable safety profiles despite higher toxicity in the combination arm.
These findings suggest that zanidatamab, whether used alone or with chemotherapy, could offer a compelling therapeutic option in a patient population with limited treatment alternatives.
