In this phase I study, combination therapy with divarasib—a next-generation KRAS G12C inhibitor—and migoprotafib—a SHP2 inhibitor—demonstrated an acceptable safety profile and promising antitumor activity in KRAS G12C-positive NSCLC patients, with a 43.8% confirmed objective response rate and a 15.2-month median progression-free survival in KRAS inhibitor–naïve patients.
The data suggest that concurrently targeting KRAS G12C and SHP2 may enhance clinical efficacy and offer a new therapeutic approach for patients with KRAS-driven NSCLC.
