In a Phase 2 trial, D3S-001—a next-generation GDP-bound KRAS G12C inhibitor engineered for rapid and complete target engagement—was administered as monotherapy to 20 NSCLC patients who progressed on prior KRAS G12C inhibitors. The study reported a 30% partial response rate, an 80% disease control rate, and marked ctDNA reduction in 11 of 14 baseline KRAS G12C–positive patients, including complete clearance in 5 of 6 radiological responders, with a tolerable safety profile.

These findings suggest that D3S-001 effectively overcomes diverse mechanisms of resistance, including KRAS amplification, offering a promising new treatment option for patients with KRAS G12C–mutant NSCLC who have exhausted earlier therapies.
